Natural killer (NK) cells are an important immune cell Group to monitor the early development of tumor, and the interaction between the activated receptor NKG2D (natural-killer Group 2 member D) on the cell surface and the ligand NKG2D on the tumor cell surface plays an important role in the process of tumor immune monitoring by NK cells. Down-regulation or deletion of the expression of NKG2D ligands will result in weakened immunogenicity of tumor cells, thereby escaping the recognition and killing effect of NK cells. Therefore, activating the expression of NKG2D ligands on the surface of tumor cell membrane is an effective strategy to enhance the immune monitoring and clearance of NK cells.

Chemical biology and molecular biology study found Parvifoline AA (PAA) through targeted peroxidase I and II (Peroxiredoxins I/II, Prxs - I/II) lead to active Oxygen levels in the cell (Reactive Oxygen Species, ROS) rise, and activate cells regulate protein kinase activity of ERK, which significantly increases the expression of NKG2D ligands, liver cell membrane surface, in turn, activate the body's immune activity of NK cells, Promote the immune killing and elimination of NK cells to liver cancer cells in vivo and in vitro. The results not only revealed the important regulatory role of prx-i /II in NK cell-mediated liver tumor immunity, but also suggested that khusane-type diterpenoid PAA, as a sensitizer for NK cell carcinoma immunotherapy, has important drug development potential and application prospect. This study provides a potential important molecular target and drug precursor for NK cell-based tumor immunotherapy.
