Alkaloids from Oxytropis ochrocephala and antiproliferative activity of sophoridine derivatives against cancer cell lines.[Pubmed: 26865176]
Ten alkaloids (1-10), with sophoridine (1) as the most abundant component, were obtained from the whole plants of Oxytropis ochrocephala Bunge. Furthermore, eight new sophoridine derivatives (11-16, 20, 21), with modification on the C-14 position of 1 were synthesized. All compounds (1-16, 20, 21) were evaluated for antiproliferative activity against five human tumor cell lines. Among them, the newly synthesized derivative 20 exhibited the best inhibitory activity against the tested cell lines. Its activity was increased by more than fourfold as compared with parent compound 1.
De novo Transcriptome Assembly of a Chinese Locoweed (Oxytropis ochrocephala) Species Provides Insights into Genes Associated with Drought, Salinity, and Cold Tolerance.[Pubmed: 26697040]
Locoweeds (toxic Oxytropis and Astraglus species), containing the toxic agent swainsonine, pose serious threats to animal husbandry on grasslands in both China and the US. Some locoweeds have evolved adaptations in order to resist various stress conditions such as drought, salt and cold. As a result they replace other plants in their communities and become an ecological problem. Currently very limited genetic information of locoweeds is available and this hinders our understanding in the molecular basis of their environmental plasticity, and the interaction between locoweeds and their symbiotic swainsonine producing endophytes. Next-generation sequencing provides a means of obtaining transcriptomic sequences in a timely manner, which is particularly useful for non-model plants. In this study, we performed transcriptome sequencing of Oxytropis ochrocephala plants followed by a de nove assembly. Our primary aim was to provide an enriched pool of genetic sequences of an Oxytropis sp. for further locoweed research.
Bacillus radicibacter sp. nov., a new bacterium isolated from root nodule of Oxytropis ochrocephala Bunge.[Pubmed: 26214748]
A Gram-positive, facultative anaerobic, rod-shaped, and endospore-forming strain, designated 53-2(T) was isolated from the root nodule of Oxytropis ochrocephala Bunge growing on Qilian mountain, China. The strain can grow at pH 7.0-8.0, 10-50 °C and tolerate up to 11% NaCl. Optimal growth occurred at pH 7.2 and 37 °C. The result of BLASTn search based on 16S rRNA gene sequence revealed that strain 53-2(T) , being closest related to Bacillus acidicola 105-2(T) , possessed remote similarity (less than 95.64%) to the species within genus Bacillus. The DNA G + C content was 37.8%. Chemotaxonomic data (major quinone is MK-7; major polar lipids are diphosphatidylglycerol, phosphatidylglycerol, unknown phospholipid, and aminoglycophospholipid; fatty acids are anteiso-C15: 0 , iso-C15:0 and anteiso-C17: 0 ) supported the affiliation of the isolate to the genus Bacillus. On the basis of physiological, phylogenetic, and biochemical properties, strain 53-2(T) represents a novel species within genus Bacillus, for which the name Bacillus radicibacter is proposed. The type strain is 53-2(T) (=DSM27302(T) =ACCC06115(T) =CCNWQLS5(T) ).
Selection of appropriate reference genes for quantitative real-time PCR in Oxytropis ochrocephala Bunge using transcriptome datasets under abiotic stress treatments.[Pubmed: 26175743]
Oxytropis ochrocephala Bunge, an indigenous locoweed species in China, poses great threats to livestock on grasslands. There is a need for further genetic study in the plants per se, for understanding the basis of its acclimation mechanism in various unfavorable environmental conditions and to implement effective control measures. Quantitative real-time reverse transcription-polymerase chain reaction (qRT-PCR) is the most commonly used method for gene expression analysis. To facilitate gene expression studies and obtain more accurate qRT-PCR data, normalization relative to stable reference genes is required. The aim of this study was to select the most stable reference genes for transcriptional analysis in O. ochrocephala.
Traffic-related trace element accumulation in roadside soils and wild grasses in the Qinghai-Tibet Plateau, China.[Pubmed: 24380977]
This research examines traffic-source trace elements accumulations and distributions in roadside soils and wild grasses in the Qinghai-Tibet Plateau. A total of 100 soil samples and 100 grass samples including Achnatherum splendens, Anaphalis nepalensis, Artemisia sphaerocephala, Carex moorcroftii, Iris lacteal, Kobresia myosuroides, Oreosolen wattii, Oxytropis ochrocephala and Stellera chamaejasme were collected at 100 sites from different road segments. The contents of metals and metalloids, including Cu, Zn, Cd, Pb, Cr, Co, Ni and As, in the soil and grass samples were analyzed using ICP-MS. The total mean concentrations of the eight trace elements in soils are Cu (22.84 mg/kg), Zn (100.56 mg/kg), Cd (0.28 mg/kg), Pb (28.75 mg/kg), Cr (36.82 mg/kg), Co (10.24 mg/kg), Ni (32.44 mg/kg) and As (21.43 mg/kg), while in grasses are Cu (9.85 mg/kg), Zn (31.47 mg/kg), Cd (0.05 mg/kg), Pb (2.06 mg/kg), Cr (14.16 mg/kg), Co (0.55 mg/kg), Ni (4.03 mg/kg) and As (1.33 mg/kg). The metal and metalloid concentrations in the nine grass species were all below the critical values of hyperaccumulators. The mean values and Multivariate Analysis of Variance (MANOVA) results indicate that: (1) the concentrations of the trace elements in the soils are higher than those in the grasses, (2) the concentrations of Cu, Zn, Cd, Pb in the soils decrease as the roadside distance increases, (3) the concentrations of trace elements in the grasses are the highest at 10 m from the road edge, (4) the higher the traffic volume, the higher the concentrations of the trace elements in the roadside soils and grasses, and (5) when the land cover is meadow, the lower the sand content in the soil, the lower the trace element concentrations. With a trace element's bioavailability represented by its transfer factor (TF) from the soil to the grass, the TFs of the eight trace elements are not in the same orders for different grass species.
Rhizobium qilianshanense sp. nov., a novel species isolated from root nodule of Oxytropis ochrocephala Bunge in China.[Pubmed: 23142858]
During a study of the diversity and phylogeny of rhizobia isolated from root nodules of Oxytropis ochrocephala grown in the northwest of China, four strains were classified in the genus Rhizobium on the basis of their 16S rRNA gene sequences. These strains have identical 16S rRNA gene sequences, which showed a mean similarity of 94.4 % with the most closely related species, Rhizobium oryzae. Analysis of recA and glnA sequences showed that these strains have less than 88.1 and 88.7 % similarity with the defined species of Rhizobium, respectively. The genetic diversity revealed by ERIC-PCR fingerprinting indicated that the isolates correspond to different strains. Strain CCNWQLS01(T) contains Q-10 as the predominant ubiquinone. The major fatty acids were identified as feature 8 (C18: 1ω7c and/or C18: 1ω6c; 67.2 %). Therefore, a novel species Rhizobium qilianshanense sp. nov. is proposed, and CCNWQLS01(T) (= ACCC 05747(T) = JCM 18337(T)) is designated as the type strain.
Seed rain and its relationship with above-ground vegetation of degraded Kobresia meadows.[Pubmed: 22688506]
Seed rain is a crucial element in vegetation regeneration, but has been rarely studied in high altitude regions, particularly degraded Kobresia meadow. Weed infestation is a distinctive feature of pasture degradation in Kobresia meadows on the Tibetan plateau, the ecological mechanism of which is closely related with vegetation's seed rain. In this paper we assess the effect of vegetation degradation on seed rain and consider its implication for restoration of degraded Kobresia meadows in the headwater area of Yellow river, through analysis of seed species composition, number of seeds landing per m(2) of soil surface, and their relationship with above ground vegetation. Vegetation degradation had an impact on the species composition and numbers of seeds in seed rain and their relationship with above-ground vegetation. Within the un-degraded meadow, which provided a closed vegetation cover, 35 % of the seed rain was of sedge and gramineae species. However, within the degraded meadows, as the extent of degradation increased, so the total number of seeds m(-2) increased, with those derived from sedge and gramineae species forming a declining proportion of the total. Degradation of Kobresia meadow on the Tibetan plateau is exacerbated by the seed input of weed species (such as Oxytropis ochrocephala, Carum carvi, Aconitum pendulum, Pedicularis kansuensis in this study). Therefore, a major priority for the restoration of such degraded meadows should be the elimination of these weeds from the above ground vegetation by human intervention.
A new flavonol from Oxytropis ochrocephala Bunge.[Pubmed: 22494026]
A new acylated flavonoid, 3-O-rhamnocitrin-6-O-benzoyl-β-D-glucopyranoside (1), together with five known flavonoids, rhamnocitrin (2), pratensein (3), (3R)-7,3'-dihydroxy-2',4'-dimethoxyisoflavan (4), (3R)-7,2'-dihydroxy-3',4'-dimethoxyisoflavan (5) and isoliquiritigenin (6), was isolated from Oxytropis ochrocephala Bunge. The structural elucidation of the isolated compounds was primarily based on HRESIMS, IR and 1-D- and 2-D-NMR analyses.
The biogeochemistry of selenium in Sunan grassland, Gansu, Northwest China, casts doubt on the belief that Marco Polo reported selenosis for the first time in history.[Pubmed: 18351293]
In order to clarify the historic academic problem of whether or not livestock poisoning in ancient Suzhou of Northwest China, recorded by Marco Polo in 1295, was selenosis, this study deals with the biogeochemistry of selenium in Sunan County in the Hexi Corridor, which is part of ancient Suzhou in China. It was found that quite a number of farm animals had suffered from intoxication and died as a result of grazing poisonous grasses, mostly Oxytropis DC, Stellera chamaejasme, and Achnatheru inebrians. Toxic symptoms of livestock grazing on Oxytropis DC are similar to those of selenium toxicity, for instance, hair loss and hoof lesions as described by Marco Polo. Therefore, we thought that toxic grass, probably Oxytropis DC, led to the intoxication of livestock recorded by Marco Polo. Average Se concentrations in two members of this species were 0.112 +/- 0.038 mg/kg for the root of Oxytropis glabra, 0.102 +/- 0.027 mg/kg for the stem and leaf of Oxytropis glabra, and 0.066 +/- 0.009 mg/kg for Oxytropis ochrocephala. The average soil selenium concentration was 0.205 +/- 0.127 mg/kg on grassland producing Oxytropis glabra and 0.152 +/- 0.024 mg/kg on grassland producing Oxytropis ochrocephala. The average Se concentration in other plants was 0.076 mg/kg in the root of Ephedra monosperma Mey, 0.029 mg/kg in the root of Rheum palmatum, 0.031 mg/kg in the root of Stellera chamaejasme, 0.037 mg/kg in Achnatherum inebrians, and 0.067 mg/kg in forage grass (Achnatherum splendens ohwi). Selenium concentrations in soils and plants in Sunan County are far less than the thresholds causing selenium toxicity in livestock. As a result, this study concludes that the livestock poisoning recorded by Marco Polo in 1295 might not have been selenosis.
