Characterization of a polysaccharide from Rosa davurica and inhibitory activity against neutrophil migration.[Pubmed: 27112979]


The rapid recruitment of neutrophils from peripheral blood into infected sites is critical step for inflammatory responses; however, the excessive and improper recruitment can lead to serious tissue damages. Thus, it is a promising strategy to inhibit their excessive recruitment for treating inflammation-related disease. Here, we isolated a polysaccharide (RDPA1) from Rosa davurica, to evaluate its physicochemical property and inhibitory effects on neutrophil migration. RDPA1 was obtained by hot-water extraction, ethanol precipitation, and fractionated by DEAE-cellulose and Sepharose CL-6B columns. RDPA1 significantly inhibited in vitro migration of human neutrophils evaluated by transwell chamber and impacted the migratory behavior observed by time-lapsed microscopy, we found the migrated distance and average velocity of RDPA1-treated cells were greatly reduced. In addition, RDPA1 treatment impaired in vivo neutrophil infiltration in the peritonitis mice. RDPA1 exhibited significant blocking capacity of the interaction between β2 integrins and ICAM-1 evaluated by flow cytometry and in vitro protein binding assay. Together, these results suggest RDPA1 could be considered as a potential candidate for developing a novel anti-inflammatory agent.


A new flavonoid glycoside and other chemical constituents from leaves of Rosa davurica and their antioxidant activity.[Pubmed: 23802510]


A 70% ethanol extract from the leaves of Rosa davurica showed significant antioxidant activity in both superoxide anion and 1,1-diphenyl-2-picrylhydrazyl (DPPH) free radical-scavenging assays. Phytochemical study of this extract resulted in the isolation of eight compounds, including a new flavonoid glycoside. The chemical structure was determined by various spectroscopic analyses. The isolated compounds and their structurally related compounds, belonging to two classes: quercetin 3-O-glycosides and gallic acid derivates, were evaluated for their superoxide anion- and DPPH free radical-scavenging activities. These compounds showed significant superoxide anion-scavenging activity with the EC50 values ranging from 1.68 to 18.09 μM, and DPPH free radical-scavenging activity with the EC50 values ranging from 7.18 to 67.62 μM. The structure-activity relationship was also reported.


The antidiabetic effects of an herbal formula composed of Alnus hirsuta, Rosa davurica, Acanthopanax senticosus and Panax schinseng in the streptozotocin-induced diabetic rats.[Pubmed: 23610602]


A folk prescription consisting of Alnus hirsuta, Rosa davurica, Acanthopanax senticosus and Panax schinseng has been used in the treatment of diabetes mellitus. The aim of the present investigation was to evaluate the antidiabetic effects of the herb formula extract (HFE) composed of Alnus hirsuta, Rosa davurica, Acanthopanax senticosus and Panax schinseng in the streptozotocin (STZ)-induced diabetic rats. The HFE was mixed in the food supply of the healthy and STZ-induced diabetic male Sprague-Dawley rats, and its effects on the body weight, water and food intake, hyperglycemia, hypolipidemic and islet structure were studied. The treatment of the rats with STZ for 6 weeks resulted in marasmus, polydipsia, polyphagia, hyperglycemia and hypoinsulinemia. In addition, the diabetic rats showed an apparent decrease in the insulin immunoreactivity and the number of β-cells in the pancreas. The addition of the HFE to the rats' food supply significantly lowered the serum glucose and the serum triglycerides level and preserved the normal histological appearance of the pancreatic islets. These results indicate that the HEF have a strong antidiabetic potential along with the significant hypoglycemic and hypolipidemic effects, which may be applicable in the pharmaceutical industry.


Anti-inflammatory, anti-angiogenic, and anti-nociceptive activities of the chloroform fraction of a methanol extract from Rosa davurica Pall. leaves in experimental animal models.[Pubmed: 20550430]


Rosa davurica Pall. has been traditionally used to treat inflammatory diseases and tumors. Its dried leaves were extracted with absolute methanol, and the methanol extract was successively fractionated into n-hexane, chloroform, ethyl acetate, n-butanol, and aqueous fractions. Anti-angiogenic and anti-nociceptive activities were determined using the chick embryo chorioallantoic membrane (CAM) assay and acetic acid-induced writhing response, respectively. Anti-inflammatory activity was evaluated using two in vivo mouse models, acetic acid-induced vascular permeability and carrageenan-induced inflammation in the air-pouch. The methanol extract gave rise to significant inhibition in the CAM angiogenesis, and showed marked 1,1-diphenyl-2-picrylhydrazyl radical scavenging activity. Among the fractions prepared from the methanol extract, the chloroform fraction exhibited highest inhibitory effect in the CAM angiogenesis. The chloroform fraction displayed anti-inflammatory activities in vascular permeability and air-pouch models. In the air-pouch model, it was able to diminish exudate volume, number of polymorphonulcear leukocytes, and nitrite content. It also showed anti-nociceptive activity in the writhing response model in mice. The leaves of R. davurica possess anti-angiogenic and related anti-inflammatory and anti-nociceptive activities, which would provide some therapeutic support on its traditional use.


[Impact of soil cadmium contamination on chlorophyll fluorescence characters and biomass accumulation of four broad-leaved tree species seedlings].[Pubmed: 16883798]


In a greenhouse pot experiment,a gradient of soil Cd concentration was established by adding 0,50,100 and 200 mg CdCl2 x kg(-1) air-dried soil, and the seedlings of one-year-old Rosa davurica, Philadelphus schrenkii, Acer mandshurica and Lonicera maackii were transplanted. The measurement of the seedlings' chlorophyll fluorescence parameters and biomass accumulation after 70 days of transplanting showed that with increasing soil Cd content, all the test tree species had a decrease of Fv/Fm, phi(PSII), qP and biomass, but an increase of qN. A. mandshurica had the greatest responses, followed by P. schrenkii and L. maackii, and R. davurica. The tolerance of these tree species to soil Cd contamination followed the order of R. davurica > P. schrenkii and L. maackii > A. mandshurica.

Study on the inhibitory effects of Korean medicinal plants and their main compounds on the 1,1-diphenyl-2-picrylhydrazyl radical.[Pubmed: 13678241]


A 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical-generating system was used to evaluate the antioxidant properties of Korean medicinal plants that have been used widely as folk medicines for several disorders, as well as compounds isolated from them. Among the Rosaceae, Rosa rugosa and Rosa davurica showed strong DPPH radical-scavenging activity. The most effective medicinal plant from families other than Rosaceae was Cedrela sinensis, followed in order by Nelumbo nucifera, Eucommia ulmoides, Zanthoxylum piperitum, Cudrania tricuspidata and Houttuynia cordata. These results serve as a good index of the free radical-scavenging activities of Korean medicinal plants. Furthermore, the polyphenols isolated from these plants, procyanidin B-3, (+)-catechin, gallic acid, methyl gallate, quercetin, quercetin-3-O-beta-D-glucoside, quercetin-3-O-beta-galactoside, quercetin-3-O-rutinose and kaempferol, exerted strong DPPH radical-scavenging activity. These results suggest that the Korean medicinal plants and the polyphenols isolated from them that exhibited effective radical-scavenging activity may be promising agents for scavenging free radicals and treating diseases associated with excess free radicals.


Inhibition of immediate-type allergic reaction by Rosa davurica Pall. in a murine model.[Pubmed: 10616960]


We studied the effect of Rosa davurica Pall. (Rosaceae) fruits (RdF) on immediate-type allergic reactions. RdF completely inhibited compound 48/80-induced systemic anaphylactic shock at the dose of 1 g/kg. When RdF was given as pretreatment, at concentrations ranging from 0.0001 to 1 g/kg, the serum histamine levels induced by compound 48/80 were reduced in a dose-dependent manner. RdF inhibited the passive cutaneous anaphylaxis activated by anti-dinitrophenyl (DNP) IgE antibody dose dependently. RdF also inhibited the histamine release induced by compound 48/80 or anti-DNP IgE from the rat peritoneal mast cells (RPMC). Moreover, RdF had a significant inhibitory effect on anti-DNP IgE-induced tumor necrosis factor-alpha production from RPMC. These results indicate that RdF may contain compounds with actions that inhibit mast cell degranulation in the rat.
