[Study on Flavonoids of Ophioglossum thermal].[Pubmed:30132647]

OBJECTIVE: To investigate the chemical components of Ophioglossum thermale,one of the original species of Chinese medicine "Yizhijian". METHODS: The air-dried whole plant material of Ophioglossum thermale was extracted by thermal reflux with 95%Et OH,then separated and purified by joint utilization of various chromatography techniques including macroporous resin,silica gel,Sephadex LH-20 column chromatography and HPLC. Their structures were elucidated on the basis of spectroscopic methods. RESULTS: Six flavonoids were isolated and identified as 3-O-methylquercetin( 1),Ophioglonol( 2),3-O-methylquercetin 7-O-beta-D-glucopyranoside( 3),3-O-methylquercetin 7-O-beta-D-glucopyraosyl-4'-O-beta-D-glucopyranoside( 4),kaempferol 3-O-( 6-O-caffeoyl)-beta-D-glucopyranosyl-( 1 -->2)-alpha-L-rhamnopyranosyl-7-O-alpha-L-rhamnopyranoside( 5) and quercetin 3-O-( 6-O-caffeoyl)-beta-D-glucopyranosyl( 1-->2)-alpha-L-rhamnopyranosyl-7-O-alpha-L-rhamnopyranoside( 6). CONCLUSION: Compounds 2 ~ 6 are isolated from this plant for the first time,compounds 5 and 6are firstly isolated from Ophioglossaceae family.
Two new peroxy fatty acids with antibacterial activity from Ophioglossum thermale Kom.[Pubmed:26742995]

Two new peroxy fatty acids, thermalic acids A (1) and B (2), together with eight known compounds, (3beta)-methyl-3-hydroxy-urs-11-en-28 oate (3), luteolin (4), quercetin (5), 3-methoxyquercetin (6), Ophioglonol (7), Ophioglonol 4'-O-alpha-D-glucopyranoside (8), pedunculosumoside B (9), syringol (10), were isolated from the herba of Ophioglossum thermale Kom. The structures of 1 and 2 were identified by HRESIMS, EIMS, 1D and 2D NMR, and electronic circular dichroism (ECD) spectra. Both two acids exhibited potential antibacterial activities against Staphylococcus aureus, Bacillus subtilis, and Escherichia coli. This is the first report of peroxy fatty acids isolated from herbaceous plants of Ophioglossaceae.

Six homoflavonoids, ophioglonin (1), ophioglonin 7-O-beta-D-glucopyranoside (2), Ophioglonol (3), Ophioglonol prenyl ether (4), Ophioglonol 4'-O-beta-D-glucopyranoside (5), and isoophioglonin 7-O-beta-D-glucopyranoside (6), together with five known compounds, quercetin, luteolin, kaempferol, 3,5,7,3',4'-pentahydroxy-8-prenylflavone, and quercetin 3-O-methyl ether, were isolated from Ophioglossum petiolatum. Their structures were elucidated by analysis of spectroscopic data and chemical evidence. Compounds 1 and quercetin 3-O-methyl ether showed slight anti-HBV surface antigen activity at 25 microM.
