Introduction of the AAL-toxin polyketide synthase gene ALT1 into FUM1-disrupted Fusarium verticillioides produces metabolites with the fumonisin methylation pattern.[Pubmed:19449880]

Fumonisins and AAL toxins(AAL Toxin TB1,AAL Toxin TB2,AAL Toxin TA2,AAL Toxin TA1,AAL Toxin TC1,AAL Toxin TC2, AAL Toxin TD1,AAL Toxin TD2,AAL Toxin TE1,AAL Toxin TE2) are polyketide-derived mycotoxins produced by several important fungal pathogens of crop plants. The toxins contain a linear, dimethylated polyketide chain, but differ in chain length and methylation positions. Here, we have isolated the major metabolites from a fumonisin null mutant, Fusarium verticillioides strain 5777, that has been transformed with the AAL Toxin TB2 polyketide synthase gene (ALT1). The results showed that the metabolites maintained the chain length and methylation pattern of fumonisins. This suggests that the timing and regioselectivity of the methyltransferase of the ALT1 PKS could be reprogrammed in this Fusarium transformant.
The presence and phytotoxicity of fumonisins and AAL-toxin in Alternaria alternata.[Pubmed:8835341]

The fumonisins (FB) and AAL Toxin TB2 are known to be produced by Fusarium moniliforme and Alternaria alternata f. sp. lycopersici when grown on rice. Alternaria alternata produced 1.3-3.1 ppm FB1 when grown on PDA for 2 weeks at 26 degrees C as determined by CD-ELISA and 0.6 ppm as determined by CF/FAB/MS. ELISA consistently yielded higher results than CF/FAB/MS. The presence of FB2, FB3 and AAL-toxin were demonstrated in spores and mycelia of A. alternata by CF/FAB/MS. The presence of AAL Toxin TB2 was further confirmed by demonstration of increased free sphingoid bases in tomato plants exposed to fungal spores and mycelia. This is the first report of AAL Toxin TB2, FB2 and FB3 in spores and mycelia and confirms the presence of FB1 in A. alternata.
