Cytotoxic activity of schisandrolic and isoschisandrolic acids involves induction of apoptosis.[Pubmed:17496413]
Chemotherapy. 2007;53(4):257-62.
Cycloartane triterpenoid Schisandrolic acid and isoSchisandrolic acid were isolated from Schisandra propinqua (Wall.) Baill (Schisandraceae). Their cytotoxicity was evaluated in several cancer cell lines and primary cultured normal mouse hepatocytes. The two triterpenoids showed moderate cytotoxic activity on all tested cell lines. Fluorescence staining and cell cycle analyses were employed to elucidate the primary mechanisms of their cytotoxicity. Our results showed that two triterpenoids exerted their cytotoxic activity via G(0)/G(1) arrest and subsequent apoptosis. Furthermore, proteolytic cleavage of poly-ADP-ribose polymerase (PARP), substrate of the caspase family, was detected and associated with apoptosis in HepG2 cells induced by the two compounds.
New lignans and cytotoxic constituents from Schisandra propinqua.[Pubmed:16491454]
Planta Med. 2006 Feb;72(2):169-74.
The CHCl (3) extract from the stems of Schisandra propinqua (Wall.) Baill exhibited significant cytotoxic activity against cancer cell lines. Subsequent fractionation procedures led to the isolation of four new dibenzocyclooctadiene lignans, propinquanins A - D, along with two known lignans, schisantherin I and heteroclitin A, and three known triterpenoids, isoSchisandrolic acid, Schisandrolic acid and schisandronic acid. Their structures, including absolute stereochemistry, were elucidated through analyses of NMR data and CD spectra. The isolated compounds were tested for their cytotoxic activity in several tumor cell lines with the MTT assay. Propinquanin B ( 2) was significantly cytotoxic (IC (50) values < 10 microM) in HL-60 and Hep-G2 tumor cell lines. Cell cycle analysis and Hoechst 33 258 staining assay suggested that one possible mechanism of cytotoxic activity by 2 might be related to the induction of apoptosis.
