A Study on the Isolation and Purification of Iso-Astragaloside IV by Macroporous Adsorptive Resin.

With the content of Isoastragaloside IV as the indicator and HPLC-ELSD as the detective method,the adsorptive characteristics and elutive parameters of resin are systematically studied.When the concentration of sample solution is 1mg·mL-1 and the ratio of the volume(ml) of sample solution and the weight(g) of resin is 3∶1,D101 macroporous resin can completely adsorb iso-Astragaloside IV.When the velocity is 5.0 BV/h,distilled water,30 % ethanol,50 % ethanol and 70 % ethanol of 4 BV is successively used to elute and the elution rate of iso-Astragaloside IV in eluent of 70 % ethanol is up to 93.14 %,which indicates that D101 macroporous resin can preferably adsorb and desorb Isoastragaloside IV and is suitable for the isolation and purification of Isoastragaloside IV and this can provide reference for the large-scaled production of Isoastragaloside IV.
