Description | 1. Variabilin, a novel inhibitor of human platelet aggregation, it is a potent RGD (Arg-Gly-Asp)-containing antagonist of the fibrinogen receptor glycoprotein IIb-IIIa (GPIIb-IIIa; αIIbβ3) and the vitronectin receptor αvβ3. 2. Variabilin is an inhibitor of human secretory and cytosolic phospholipase A2 (PLA2) activities that controls eicosanoid production in vitro and in vivo, inhibits neutrophil degranulation and superoxide generation in vitro and shows anti-inflammatory activity after topical or p.o. administration to mice. 3. Variabilin is an antimicrobial furanosesterterpene. 4. Variabilin is an important chemotaxonomic marker for the family Irciniidae. |

Larvicidal activity of isoflavonoids from Muellera frutescens extracts against Aedes aegypti.[Pubmed:23156998]

The biological activity of extracts from the leaves, bark and roots of Muellera frutescens, an Amazonian ichtyotoxic plant, were evaluated to find new environmentally safe insecticides. The n-hexane extracts of bark, leaf, and root showed a strong toxic activity against Aedes aegypti mosquito larvae. Bioguided fractionation of the bark extract led to the isolation of seven isoflavonoids (12a-hydroxyelliptone, elliptone, (-)-Variabilin, rotenone, rotenolone, tephrosin and deguelin). Rotenone and deguelin are responsible for the larvicidal activity of the plant. M frutescens leaves contain up to 0.6%, w/w, deguelin. These results justify the traditional ichtyotoxic use of M frutescens. The leaves contain a relatively high proportion of deguelin and, therefore, can be considered as a renewable source of this environmentally friendly insecticidal isoflavonoid.

Total syntheses of (-)-Variabilin and (-)-glycinol have been accomplished, using the catalytic, asymmetric "interrupted" Feist-Benary reaction (IFB) as the key transformation to introduce both stereogenic centers. A monoquinidine pyrimidinyl ether catalyst affords the IFB products in over 90% ee in both cases. Other key steps include an intramolecular Buchwald-Hartwig coupling and a nickel-catalyzed aryl tosylate reduction.
