Optimized extraction process of formula granules for seeds from Desurainia sophia fried without additional ingredients by Box-Behnken response surface method.[Reference: WebLink]

To optimize extraction process of formula granules for seeds from Desurainia sophia fried without additional ingredients by the Box-Behnken response surface method.
METHODS AND RESULTS:
Several influentialfactors of extraction process were investigated by single factor experiment using UV-spectrometry,with ultrasonic power,time,temperature,and ratio of water as evaluating indexes. The conditions of the extraction were evaluated by total flavonoids of seeds from D. sophia fried without additional ingredients. Based on single factor test,three parameters were chosen to optimize the extraction process. The effects on the yield of flavonoids were analyzed by the response surface methodology,and experiments designed by the Box-Behnken method. Finally,the cream rates as well as contents of Quercetin 3-O-beta-D-glucose-7-O-beta-D-gentiobiosiden and sinapine thiocyanate were determined by HPLC as evalluation indexes. The software of Design-Expert 8. 0. 5 was used to statistically analyze the optimal extraction process. The optimal time,temperature,power,and water ratio predicted by response surface test and quadratic regression model were: 70 ℃,400 W,and 34 m L·g~(-1).
CONCLUSIONS:
The optimum extraction process of formula granules for the seeds from D. sophia fried without additional ingredients is reliable,and its precision is high,which provides a reference to draft national standards and industry standards.
